Though clearly not required for hemocyte developmental migrations, we wanted to test whether PI3K was required for hemocytes to polarize and chemotax toward a wound site. To test the involvement of PI3K in this process, we made laser ablations in embryos expressing Dp110D954A in the hemocytes. In contrast to the wild-type chemotactic response Fig. 8 A ; , the mutant cells failed to chemotax toward the wound site and the wound remained largely undetected by the hemocytes up to 1 after laser ablation Fig. 8 B ; . The same result was obtained when beads were implanted in these embryos Fig. 8 D ; . ensure that this phenotype was due to a failure in chemotaxis and not an indirect result of a reduction in hemocyte survival, we counted the number of hemocytes on the ventral side of embryos expressing Dp110D954A. We found no significant difference in hemocyte number when compared with wild-type embryos mean number, 49 9 [n 17] in Dp110D954A and 56 8 [n 25] in wild type ; , demonstrating that PI3K plays no significant role in hemocyte survival at this stage of development. This is consistent with previously published data showing that hemocyte-specific expression of Dp110D954A does not cause the hemocyte aggregation that is associated with a decrease in cell survival Bruckner et al., 2004 ; . To further verify our chemotaxis result, we implanted two beads into pxnGAL4 UAS-GFP embryos in which the hemocytes were expressing GFP but were otherwise wild type. In these experiments, one bead was presoaked in the PI3K inhibitor drug LY294002 Vanhaesebroeck.

Patients and methods: Thirty-two patients treated with cisplatin- and doxorubicin-containing chemotherapy 13 years before the time of analyses were evaluated for neuro-, oto-, pulmonary-, vascularand gonadal toxicity including evaluation of myocardial damage and cardiovascular risk factors and analysis of microcirculation. Results: Thirty percent of the patients showed abnormal left ventricle function. Elevated follicle stimulating hormone FSH ; and luteinising hormone LH ; levels in 75% of patients were often associated with low testosterone levels. Elevated total cholesterol levels were found in 82% and higher triglyceride levels in 44% of patients, most of them were overweight. About 25% of the patients developed diastolic arterial hypertension after chemotherapy. Reduced hearing was confirmed in 23% of patients, especially at frequencies higher than 3000 Hz. Moreover, 53% of patients presented transient evoked otoacoustic emissions. In 38% of patients non-symptomatic neuropathy was detected, in 28% symptomatic neuropathy, and in 6% disabling polyneuropathy. In 80% of patients with neuropathic symptoms additional morphological and functional abnormalities were found by nailfold capillary videomicroscopy, compared to only 57% of the patients without neuropathic symptoms. Conclusions: Patients cured by cisplatin-based chemotherapy for metastatic testicular cancer have to be cognizant of their unfavorable cardiovascular risk profile, that might be a greater risk than developing a relapse or second malignancy. EXPERIMENTAL PROCEDURES Materials - Coagulogen 7-9 ; and the clotting enzyme 12 ; were prepared from hemocyte lysates of Tachypleus tridentatus as described. Homobifunctional cross-linkers, disuccinimidyl suberate DSS ; , disuccinimidyl glutarate DSG ; , and disuccinimidyl tartrate DST ; were obtained from Pierce Chemical, Rockford, IL. TPCK-trypsin was obtained from Worthington Biochemical Corporation, Freehold, NJ. Cross-linking - Coagulogen 1 mg ml in 20 mM HEPES, pH 8.0 ; was incubated with the clotting enzyme at 37 C for 1 h enzyme substrate 1 38, w w ; . The resulting coagulin gel was dissolved by 10-fold dilution with the same buffer to a final concentration of 0.1 mg ml, and no clot formation was observed under these conditions. A cross-linker in dimethylformamide was added to the coagulin solution to give a final concentration of 0.2 mM, and the solution was further incubated at 25 C for 30 min. The reaction was stopped by adding 1 M Tris-HCl, pH 8.0, to give a final concentration of 0.1 M, followed by incubation at 25 C for 15 min. Cross-linked coagulin was treated with 10% trichloroacetic acid and the resulting precipitate was used for SDS-PAGE and tryptic digestion. As a negative control, cross-linking experiments for coagulogen were carried out under the same conditions. SDS-PAGE was performed in 15% slab gels under reducing conditions, according to the procedure described by Laemmli 13 ; . The gels were stained with Coomassie brilliant blue R-250. Acid- or alkali-treated plasma was exposed to acid pH 1 ; or alkali pH 11 ; for one hour at 4C and neutralized. Heat-treated plasma was heated to 90C for 15 min. In the initial stage of aggregation, hemocytes containing many vacuoles with filament-like inclusions hemocytes of type C named by Azumi et al., 1993 ; make contact to form small homogeneous aggregates. At the same time, amoeba-like hemocytes type A and B hemocytes ; migrate toward and contact vacuolated cells containing several vacuoles of high density type F hemocytes this triggers and promotes the aggregation of the latter type F hemocytes themselves. Finally, several large heterogeneous aggregates including almost all types of hemocytes are formed. Six types were identified: prohemocytes PR ; , plasmatocytes PL ; , granulocytes GR ; , spherulocytes SP ; , adipohemocytes AD ; , oenocytoids OE ; , with two additional types, vermicytes VE ; and podocytes PO ; , having debatable identities. The PRs were mostly rounded small cells with characteristically large and filling nucleus showing an uneven surface due to the presence of vaguely concentric and elevated circles by SEM. The PLs were polymorphic but largely spindleshaped. The GRs were rounded to ovoid with relatively small and eccentric nucleus. The SPs were also rounded to ovoid with cytoplasm appearing lobulated. The ADs were rounded to elongate with cytoplasm containing lipid droplets. The OEs were rounded, and mostly large. The PL and GR remained relatively high and SP, AD and OE low throughout the postembryonic development. The VEs, were greatly elongated and narrow cells with pointed ends. The POs were flat cells with three or more cytoplasmic arms projecting from a central body. These later cells appear for a limited period only in the early pupa. The THC steadily decreases during the developmental stages, attains its peak in the late 5th prepupa ; instar and steeply declines in the pupa.
