As ubiquitination seems to be an important regulatory mechanism for receptor-trafficking, several studies need to be performed in order to demonstrate what sort of signals and processes i.e receptor agonists, exposure to hormones, altered levels of neuron activity ; shown previously to modify surface expression or density of AMPA or NMDA receptors, are mediated by ubiquitination. Moreover, questions arise whether such molecular processes are significantly important in specific forms of synaptic plasticity such as LTP or LTD Turrigiano, 2002 ; . Overall, this set of results demonstrate that cellular expression of the AMPAGLR-1 receptor subtype at synapses is regulated by two specific intracellular processes: one, the clathrinmediated endocytosis and an other which is mediated by ubiquitination Turrigiano, 2002 ; . Recent experiments have shown that ZNRF proteins constitute a family of presynaptic E3 ubiquitin ligases, where ZNRF1 and ZNRF2 are proteins that contain an E# ubiquitin ligase activity that is highly expressed in the CNS of mammals, particularly active during development Araki, Milbrandt, 2003 ; . These proteins have been shown to be located in different compartments within presynaptic terminals, where ZNRF1 is associated with synaptic vesicle membranes, whereas ZNRF2 is highly expressed in presynaptic membranes Araki, Milbrandt, 2003 ; . Experiments inducing mutant protein ZNRF species, disrupting the C-terminal domain which contains a RINGER finger structure necessary for the normal functioning of E3. The phase III AIM study confirmed and extended the findings of a phase IIb study in a similar patient population 8, 9 ; by demonstrating that abatacept, with background methotrexate, is effective in reducing the signs and symptoms of rheumatoid arthritis and improving physical function and health-related quality of life. The study also demonstrated that abatacept slows the progression of structural damage in patients with moderate-to-severe disease and an inadequate response to methotrexate treatment. We observed a rapid and sustained increase in all ACR responses through 1 year with abatacept. Both pain and self-assessment of disease activity statistically significantly improved in abatacept-treated patients, as early as day 15 in some patients. A total of 28.8% of abatacept-treated patients exhibited an ACR 70 response at 1 year, and a statistically significant number of these patients maintained their ACR 70 response for 6 and 9 consecutive months. Statistically significant proportions of patients in the abatacept group had DAS28 less than 2.6 or 3.2 or less at 6 months, and scores continued to increase through 1 year. The increasing efficacy observed during 1 year of abatacept treatment was not due to the allowance of additional medications during the second half of the study. Few abatacept-treated patients received additional disease-modifying antirheumatic drugs 3.7% for abatacept vs. 14.4% for placebo ; during months 7 through 12 of treatment. Abstract We hypothesized that suppression of endogenous testosterone inhibits the adaptations to strength training in otherwise healthy men. Twenty-two young men with minor experience with strength training participated in this randomized, placebo-controlled, and double-blinded intervention study. The subjects were randomized to treatment with the GnRH-analogue goserelin 3.6 mg ; or placebo saline ; subcutaneously every 4 weeks for a period of 12 weeks. The strength training period of 8 weeks starting at week 4 included exercises for all major muscles 3-4 sets per exercise x 610 repetitions with corresponding 6-10 repetition maximum RM ; loads, 3 week ; . A strength test, blood sampling, and whole body DXA scan were performed at weeks 4 and 12. Endogenous testosterone decreased significantly p 0.01 ; in the goserelin group from 22.6 5.5 mean SD ; nmol l to 2.0 0.5 week 4 ; and 1.1 0.6 nmol l week 12 ; , whereas it remained constant in the placebo group. The goserelin group showed no changes in isometric knee extension strength after training, whereas the placebo group increased from 240.2 41.3 to 264.1 35.3 Nm p 0.05 within and p 0.05 between groups ; . Lean mass of the legs increased 0.37 0.13 kg and 0.57 0.30 kg in the goserelin and placebo group, respectively p 0.05 within and p 0.05 between groups ; . Body fat mass increased 1.4 1.0 kg and decreased 0.6 1.2 kg in the goserelin and placebo group, respectively p 0.05 within and between groups ; . We conclude that endogenous testosterone is of paramount importance to the adaptation to strength training.
