When bound to D50S, both Synercid components interact with each other via the nucleotide A2062, which plays a key role in chloramphenicol 38, 58 ; and tylosin 24 ; binding. These interactions seem to stabilize the U2585 flipped conformation and consequently enhance the drug antimicrobial activity. Importantly, U2585 conformation is hardly changed in an H50S-SA complex 25 ; , indicating a different binding mode Figure 3g ; consistent with the inherent differences in PTC conformations between H50S and eubacterial ribosomes 28, 85 ; . Similarly, the SB interactions with A2058 in D50S explain why SB components do not bind to H50S 25 ; , which possesses the typical eukaryotic G rather than the common eubacterial A in this position. At its unique binding orientation, TAO hits an element of the tunnel wall, the tip of protein L22 beta-hairpin, a tunnel constituent reaching from close to the PTC to the vicinity of the tunnel opening on the other side of the large subunit 28, 46 ; . US: From an increase in construction material costs to a decrease in ships that can accommodate deep-keeled recreational craft, low water levels are causing headaches on the Great Lakes. "It has been this way for several years, but it is particularly problematic this spring. The water level is lower, " said Duncan Henderson, executive director of the Waukegan Port Authority, Illinois. Henderson added that the problem affects both large commercial ships delivering cement and gypsum to Lafarge and National Gypsum on Waukegan's lakefront, and recreational boats in Waukegan harbour. Henderson noted that Lafarge and National Gypsum provide construction materials for not just Lake County but a sprawling area stretching into Wisconsin and Indiana. When the companies have to run more ships to deliver the same amount of materials because of weight limitations, he said, those rising costs will be passed along. Shippers are forced to lighten their loads so they can move safely into harbours. The lighter loads sometimes hundreds of tons per ship turn into headaches for suppliers that send their goods on vessels, shippers and companies whose orders come up short. Midwest Energy Resources, a coal supplier, sent out its first vessel of the season 2007 with a load just under 60, 000t, shy of a typical 62, 000t shipment. For every inch the lakes recede, ships must reduce their loads between 50 and 270t, Henderson explained. At the end of the 2006 season, with waters particularly low on Lake Superior, ships lost about 8000t per trip about 11% of their carrying capacity, said Glen Nekvasil, vice president of corporate communications for the Lake Carriers' Association, a trade group for shippers on the Great Lakes. "Every ton has an impact. These companies, they earn their living carrying cargo, so every lost ton of cargo is lost revenue, " Nekvasil explained. Water levels have slipped for years and the forecast isn't getting any better. Lake Superior is within a few inches of its record low 599.5ft deep, set in 1926. It's now about a foot lower than in 2006 and projections for this summer are that it'll get even lower. When large areas of the lakes do not freeze over, evaporation becomes a year-round factor and lowers lake levels. "It does cut back on how much tonnage we can run into Waukegan harbour, " said Larry Brewer, manager of Lafarge's Waukegan operations. Brewer said that the impact has been minimal so far, but the shipping season has just started. "It will affect us eventually, " he said. So far, Lafarge has had three cement vessels in Waukegan Harbour in spring 2007. National Gypsum welcomed its first ship in April 2007, and Waukegan plant manager Steve Rogers said that he is still assessing the impact of lower water levels and expects to know more in a week or two. "If we have to lower our tonnage, it will be more expensive, " he said. Henderson said that dredging alone will not solve the problem, and beyond that, there isn't much that can be done. Animal data identify involvement of CYP's 2B and 3A family enzymes in PCB metabolism and inductive effects of PCBs metabolites. Minimal information is available on human CYP 450 isoform specific metabolism of PCBs. Experiments were designed to study the metabolism of PCB 101 by human isoforms of CYP 450. Ten pmol of different Gentest Supersomes baculovirus transfected insect cell microsomes expressing specific human CYP 450s isoforms ; containing human CYP 2A6, 2B6, 3A4, and 2D6, were incubated separately with 30mcM PCB101, and an NADPH regeneration system. CYP 2A6 was the only isoform that produced metabolites. The major quantifiable metabolite was identified as 4-OH PCB101. Product formation followed Michaelis Menton kinetics. Trace amounts of 3-OH were present but not quantifiable. The km for CYP 2A6 conversion of PCB101 to 4-OH PCB101 was 36.4mcM with a Vmax of 0.53pmol min pmol CYP 2A6. PCB101 hydroxylase activity in human liver microsomes HLM ; matched HLM content of CYP 2A6. These data support CYP2A6 as the specific isoform responsible for PCB101 metabolism in humans. This is in contrast to the 2B and 3A isoforms identified in animal studies.
